چکیده

The enzymatic hydrolysis of urea by jack bean urease was investigated at 25 °C over the pH range 4–9. Reaction rate data were found to be well described by a modified Michaelis-Menten equation with a pH-dependent rate coefficient and a product inhibition term. The influence of pH on activity was interpreted in terms of perturbation of the enzyme distribution among three differently protonated forms. Kinetic analysis yielded a Michaelis constant of 3.21 mmol l and indicated that the inhibition mechanism was of the fully non-competitive type, with K P = 12.2 mmol l. The estimated activation energy was 35.3 kJ mol. The resulting kinetic expression was tested by comparing model predictions with the experimental behaviour observed in unbuffered media and over a long-term period.

Kinetic Modeling of Enzymatic Hydrolysis of Pretreated Sorghum Bicolor and Rice Husk

In this study, the hydrolysis of pretreated sorghum stem and rice husk was investigated at various initial enzyme concentrations and substrate loadings. The slowdown in enzymatic hydrolysis of lignocellulosic materials with conversion has often been attributed to decreasing the activity of enzyme. A kinetic model was developed and expressed mathematically based on enzyme deactivation for enzyma...

A novel approach for stabilizing fresh urine by calcium hydroxide addition.

In this study, we investigated the prevention of enzymatic urea hydrolysis in fresh urine by increasing the pH with calcium hydroxide (Ca(OH)2) powder. The amount of Ca(OH)2 dissolving in fresh urine depends significantly on the composition of the urine. The different urine compositions used in our simulations showed that between 4.3 and 5.8 g Ca(OH)2 dissolved in 1 L of urine at 25 °C. At this...

Optimum Hydrolysis Conditions of Cassava Starch for Glucose Production

Acid and enzymatic hydrolysis of cassava starch to glucose (fermentable sugar) were investigated and compared. And the effects of acid concentration, pH, temperature and time on the yield of glucose were studied. Experiments were carried out at a temperature range of (60 – 100)C between 30 minutes and 4 hours. (0.2 – 1.0)M strength of H2SO4 acid was used and pH values range of 4 – 7 was conside...

Reaction pathways and free energy profiles for spontaneous hydrolysis of urea and tetramethylurea: unexpected substituent effects.

It has been difficult to directly measure the spontaneous hydrolysis rate of urea and, thus, 1,1,3,3-tetramethylurea (Me4U) was used as a model to determine the "experimental" rate constant for urea hydrolysis. The use of Me4U was based on an assumption that the rate of urea hydrolysis should be 2.8 times that of Me4U hydrolysis because the rate of acetamide hydrolysis is 2.8 times that of N,N-...

Whole cell kinetics of ureolysis by Sporosarcina pasteurii.

AIMS Ureolysis drives microbially induced calcium carbonate precipitation (MICP). MICP models typically employ simplified urea hydrolysis kinetics that do not account for cell density, pH effect or product inhibition. Here, ureolysis rate studies with whole cells of Sporosarcina pasteurii aimed to determine the relationship between ureolysis rate and concentrations of (i) urea, (ii) cells, (iii...
